Biology Microarray Lab report Essay

The analysis of DNA using the microarray technique has become one of the most significant methods in the area of research genetics. This technique falls under the area of gene expression profiling. Most of the time, this procedure is applied by scientists in the effort to investigate a wide range of conditions. This is because experimental procedures cam be performed on numerous genes at the same time. They include researches on cancer to finding numerous solutions to the problems that are presented by pests. With this advancement an opportunity has been offered for the performance of personal DNA microarray experiments. Among the basis of such experiments is the determination between healthy cells and the cancer cells. Based on the complexity of the microarray experiments, it is vital that all scientists obtain a solid understanding on the DNA basics as well as the way through which genes express themselves. DNA microarrays have been used in the extensive survey of the relative transcription in any gene within a genome. Most of the cancer cells in human beings are found within the developing nerves. Howevber, they do not allow the complete quantification level of gene expression. Moreover, the DNA chips do not make it possible to determine the amount of mRNA produced from a relative sample with that produced from the control population. As such, it can be used to compare the rate of gene expression in a lung cell with cancer and a helthy lung cell. Therefore, the main goal of this practival test is to ptovide a way to understand how microarrays are used tostudy the gene expressions. It allows the investigators to determine the level of gene activity for a complete gene. As such, they make it easier to diagnose various diseases that injmclude cancer. Two main steps will be involved in the performance of the microarray lab experiments. These include the prehybridization and the hybridization steps. These are conducted through a number of 7 mini steps. They will involve the collection of the tissue or sample, the isolation of the RNA, isolation of the mRNA, creation of a labelled DNA copy, application off DNA, scanning of a microarray and the analysis of data (Campbell et al., 333). Different pH indicators that are colorless at neutral and colored at high pH of above 10 will be applied. They will be mixed with molten Agarose; this includes Madison, Promega, WI and V312A. It will later be allowed to cool. They could also be placed in a hot bath of 650 and kept molten. They will be melted if to be used days later. Pipettes will be used to apply the DNA onto the slides. Collection of mRNA The plate will be incubated for 5 minutes to allow for the release of mRNA. It will then pipetted in a Tri reagent for extraction. 80 uL of chloroform will subsequently be added and shaken vigorously then centrifuged to separate the cells into layers. 2 ml isopropanol will be added, the mixture centrifuged and the supernatant poured off. After this, the preparation of the RNA for spec by will be done by adding Agarose gel. The pre-hybridization steps will involve the preparation of stocks and obtaining of the microarray slide and steaming it on a hot plate for between 30 seconds and 1 minute. It will then be cooled at room temperature. It is important to warm the solution in case there are any crystals. The two slides can then be treated back to back and dipped in distilled water severally; it will be dried and spun for 2-3 minutes in a centrifuge. The slide is then hybridized by placing in a clean 50ml tube in a heated incubator. A coverslip is prepared by dipping into 0.2%SDS, then water. Blot, dry and continue to the hybridization step (Campbell et al., 338). Hybridization It includes the hybridization of the DNA chip using 3DNA array 350 protocol. Chips containing 70mer oligos and 2 copies of the known cDNAs in the human genome are used. This should be done at least 24 hours before the experiment. Make the solution only when it is ready for use. It is mainly 0.1M NaOH. The first step includes thawing vial at 7.2X. Make the hybridization solution with 50 ul total to fit across the cover slip. Incubate it at 800 for ten minutes. The entire 58 ul is then transferred on the microarray and the short edge of the cover slip placed on the short edge of the slide, which is then transferred to a 50 ml tube. The brand Rasna is owned by Pioma Industries that introduced the concept of Soft Drink Concentrate (SDC), a segment that had been created and nurtured by the company in the Indian beverages Market. The brand was originally launched by the name of ‘Jaffe’ in 1976 and marketed with the help of Voltas, but was renamed to ‘Rasna’ in 1979. When the product did not do as well as it was expected, in 1983, the advertising was handed over to an advertising agency named Mudra Communications Ltd. Pioma Industries Limited is perhaps not a familiar name for the average Indian consumer. But Pioma’s brand Rasna is very well known. In fact, the name Rasna is almost a generic name for soft drink concentrates in Indian beverages market. Rasna’s extremely popular advertisements with the tagline â€Å"I love you Rasna† had become an integral part of the Indian advertising folklore. In March, 2002, Pioma announced a radical overhauling of its strategies for the Rasna Brand. This development was rather unexpected, as the brand had been lying dormant since long. Rasna is the market leader in the 250 Crore Indian soft drink concentrate industry. The SDC industry is miniscule compared to the 5000 crore carbonated soft drink industry (CSD). Try to minimise waste in production, so that the cost of production comes down further. 2. Venture into new products, like ready to drink bottled drinks. 3. A Doll’s House – Henrik Ibsen

A Doll's House, by Henrik Ibsen, is a well written play portraying women's struggle for independence and security in the nineteenth century. The drama revolves around Nora, a traditional housewife, who struggles to find a way to save her husband's life while battling society's norms. Her decision to forge a check to help her husband, Torvald, went against the patriarchical laws of that time: her plan was to borrow money and save her ill husband's life. Krogstad, the antagonist, discovers her deceit and threatens to expose her lies. As a result, the protagonist is torn between telling the truth to her husband or attempting to cover it up to save her marriage. In the following analysis, I'll discuss Torvald and Nora's relationship and Torvald's general view of women. In addition, I will evaluate his attraction to Nora, focusing on his attitude on treating like a doll, or a child. Lastly, I'll discuss how and why Torvald spoils Nora by giving her what she wants. Torvald and Nora's relationship appears to be more of a father-daughter relationship rather than a traditional husband-wife relationship. He spoils her, and further, finds reason to rain affection on her: â€Å"He goes up to her and takes her playfully by the ear. † Torvald views the effeminate, dainty Nora as a child, and refuses to take her seriously. His view was very much like male dominant view of women in that era: women were innocent, naiive, little â€Å"skylarks† and â€Å"squirrels† who were helpless and vulnerable. Torvald also believes the measure of a women is her man and he reinforces that view when he toys with Nora, â€Å"There you are. Gives her money] Do you think I don't know what a lot is wanted for housekeeping at Christmas-time? † This example illustrates his control over Nora. Despite her asking for money, Torvald playfully lectures her, but ultimately hands her the money and fulfills her desires. His playfulness is another facet of the dominant male symbol, playing games with a women's mind to prove his manly independence. There are other instances where Torvald reinf orces the patriarchical view of European society. For example, when Nora and Torvald were discussing money and loans, Nora mentions that she wouldn't bother with people who lend her money. Torvald replies, â€Å"That is like a woman! † His statement is key to understanding their relationship. Nora reinforces the husband-daughter paradigm by playing the role of a naiive wife, which Torvald easily falls for. And when Nora brings up Krogstad, Torvald is even more surprised by her comments: â€Å"Nora, Nora, and you would be a party to that sort of thing? And to tell me a lie into the bargain? Torvald believes that Nora should be under his influence; thus, a dominant male's influence. His idealistic view of women reinforces his manly independence persona towards Nora and proves that he is of touch with play's reality. He also states his opinions about the upbringing of â€Å"morally corrupted† people. Torvald lays the blame on their mothers saying, â€Å"Almost everyone who has gone to the bad early in life has had a dec eitful mother. † Torvald is close minded and believes strongly that morally corrupt is due to mistakes of a female's (mother's) upbringing. Simultaneously, Torvald is attracted to Nora, because she gives him all the credit and feeds his ego. He strongly believes that without him, she would not be able to live her life. When Nora asks Torvald to go to the fancy-dress ball, he replies,† Aha! So my obstinate little woman is obliged to get someone to come to her rescue? â€Å". Torvald enjoys being her hero, which reinforces his manly independence. Time and again, he is there to ‘save' her. To be clear, Nora does not stop Torvald possessiveness over her, explaining â€Å"Torvald is so absurdly fond of me that he wants me absolutely to himself. Nora is an object he owns and controls, while Nora continues to take her role as damsel-in-distress. Nora plays perfectly the role of a young doll in a play house. Torvald treats her like a child, which allows her to exploit him. She continually manipulates him for her desires. When Nora, â€Å"[Smiling quietly and happily] You haven't any idea how many expenses we skyla rks and squirrels have, Torvald. † Whether its societal pressure or her own views, Nora allows herself to be treated like a child and allows her husband to feel as if he's in control of their elationship. She admits that she acts the way she is, because she thinks of, â€Å"How painful and humiliating for Torvald, with his manly independence; to know that he owed me anything! It would upset our mutual relationship†. Its ironic to realize that their entire relationship is based on lies. Torvald's main priority is to thrust his manly independence and power over Nora, as both play the part of actors in society's drama. Torvald challenges Nora's attitude and character in the hope of having her fit his idealistic view of women. Torvald wants an out-of-touch-with-reality Nora in his life. As Nora explains, â€Å"Torvald can't bear to see dress making going on. † Torvald does not want to see Nora as an independent and rebellious woman. Her true self is quite different to what she portrays, yet she continuously plays this character that Torvald will agree with. Torvald later forgives Nora for bringing up the topic of Krogstad employment: â€Å"because it is such eloquent witness to your great love for me. † This clearly shows that their relationship is ruled by the more powerful character. Torvald wants Nora to be this weak and submissive woman by spoiling her so that he ends up reinforcing his manly independence. He even says,†I am man enough to take everything upon myself. † He is taking credit from their relationship, and doesn't give Nora any credit, because she's a woman. She is only there to compliment him and feed his ego. However, Torvald is concerned when he sees Nora all worn out, â€Å"But dear Nora, you look so worn out. Have you been practicing too much? † Here, Torvald thinks that Nora's worn out, because of practicing the tambourine. The truth is that Nora is worn out because of practicing her character to suit Torvald's life and attempting to save her marriage. Ibsen's A Doll House proves to be a critique of the traditional roles of men and women in 19th Century marriage. Nora treated by her father as a doll-like child, continues to be treated by her husband in the same way. This is how we begin to understand how Torvald spoils Nora as an act of reinforcement of his manly independence rather than an act of love.

It is the process of creating structure, establishing relationships, and allocating resources to accomplish the goals of the organization. Directing involves the guiding, leading, and overseeing of employees to achieve organizational goals. Controlling involves verifying that actual performance matches the plan. If performance results do not match the plan, corrective action is taken. Henri Fayol, the father of the school of Systematic Management, was motivated to create a theoretical foundation for a managerial educational program based on his experience as a successful managing director of a mining company. In his day, managers had no formal training and he observed that the increasing complexity of organizations would require more professional management. Fayol's legacy is his generic Principles of Management. Of Fayol's six generic activities for industrial undertakings (technical, commercial, financial, security, accounting, managerial), the most important were The Five Functions of Management that focused on the key relationships between personnel and its management. The Five Functions are: Planning: Drawing up plans of actions that combine unity, continuity, flexibility and precision given the organization's resources, type and significance of work and future trends. The individual chosen (D.C.) is a mentor, who is handing down the baton to me, as the clinical instructor for a group of nursing students at Biola University. D.C. has numerous years experience as a bedside nurse as well as an educator. Overview of Career D.C. always knew she wanted to be a nurse and began nursing school at the young age of 17. In 1977, she received her Advanced Degree in Nursing (ADN) from Umpqua Community College in Roseburg. She went on to receive her Bachelor’s Degree in Management from Georgefox University, in Newburg, Oregon. The positions she held included: working as a bedside nurse for three years, a nurse in critical care for seven years and a supervisor/manager for 20 years. This 30-year experience took place at a community hospital called Providence Health and Services, in Oregon. Graduate Education D.C. went on to receiver her Master’s in Leadership and Nursing Education from Warner Pacific University in Portland, Oregon, from 2006-2008. While attending graduate school she continued to work at Providence Health and Services. Her current position made it necessary to go back to school because the institution was requiring all supervisors to have a Master’s degree. D.C. enjoyed her on-line educational experience from Warner Pacific. After her degree, she became the manager/director for  multi-services including: wound care, care management, social  services, medical-surgical and telemetry units. This took place over a four-year period and then D.C. became the Director of Education at Providence for an additional three years. This is where she taught for the University of Great Falls, Montana for their RN to BSN satellite program. D.C. taught live, virtual classrooms with students from many states. She retired from Providence in 2011 and moved to California with her beloved husband, who became the Dean of Rosemead, at Biola University, in La Mirada, California. Present Position Soon after her move to California, D.C. was asked by the nursing department of Biola University, if she would consider teaching for them. She said no at first but then was enticed to begin as a part time clinical instructor for the Bachelor’s in Nursing Program. She began as a community health instructor and then applied for the full-time position as an Associate Professor. She taught Management and Leadership in the fall and Nursing Community in the spring. D.C. is a Level Coordinator for all level 3 activities in the nursing department and part of the administration team as well. Competencies learned in the graduate program included team building, coaching, developing unity and administration skills. Some of her various certifications over the years include: Medical/Surgical Certified, ACLS, PALS and NRP. She also is a master trainer for crucial conversations and a senior facilitator for select interview training. While at Biola University, D.C. also took 80 hours of epidemi ology online through the Center’s for Disease Control. She is also part of the FEMA disaster response program and held an earthquake preparedness day at the university this year. Her graduate degree gave her all the necessary tools to confidently teach at a university level. Pearls of Wisdom For any student in a graduate program it is essential to have a mentor who can give you advice. Words of wisdom from D.C. included â€Å"Everything is a conversation. Even if the conversation is  painful, be open and honest.† The next golden nugget from D.C. was, â€Å"Just do what’s next and pretty soon you’re done.† This helped me to realize to take one step or one assignment at a time and not look with fear to the future. D.C learned organizational skills and was able to write specific objectives for nursing education. Her thesis was on how to be a new graduate with confidence and competence. She learned a new appreciation for people cultural diversity. One thing she learned the most about herself was that she was biased. She encouraged me to question my biases and assumptions in everything we do. These were some incredible tools I will take with me as I continue through the graduate program. Conclusion The effects of graduate education can enhance your skills, increase your knowledge, give you more awareness culturally, enable you to have better organization and make decisions, as well as become a better communicator. D.C. felt she was able to practice her new skills at the hospital she was already working for in a safe environment with people she trusted. What was affected the most in D.C.’s experience during grad school was her attitude. She gained a broader perspective and realized there was so much to gain from others. She felt her graduate degree gave her critical thinking skills that she can use forever. D.C. was an excellent example of someone who has used their graduate degree to the fullest extent as teacher, mentor, advisor and facilitator. After approximately two minutes of breaking down the grease, the dishes were raised to a uniform height at one end and the broken down grease was allowed to run. Record your observations. Dispose of the acetone mixture in the appropriate Laboratory Byproducts Jar. Discard the polystyrene in the appropriate Laboratory Byproducts Jar.Condensation Polymer?Glottal Resin The following procedure must be carried out in a fume hood! Obtain a paper cup. Place 5 Goff ophthalmic anhy dride and 0. 25 g of sodium acetate in a disposable 18 x 150 mm test tube (provided by your TA). Add 2 ml of glycerol. Using our test tube clamp, carefully heat the mixture over a Bunsen burner flame (see TECH I. D). Heat the top of the contents first and work toward the bottom as the mixture melts. Move the test tube in and out of the flame as you do this. Note when the mixture begins to boil and then continue to heat for 3 to 4 minutes.The solution Pour the solution into the paper cup. Test the solubility of the polymer as you did for polystyrene. 5 Modification of polymers Experiment on crosslink with Glue It is recommended that gloves not be worn when working with Elm's glue. It is easier to remove glue residue from your skin than from the gloves. Obtain a plastic cup and pour 15 ml of Elm's white g lue into it.Add 15 ml of water and stir well. Record these observations. Polymer fabrication Polyethylene bottle Heat one end of a 10-15 CM piece of polyethylene tubing in a Bunsen burner flame. When the polymer is in the molten state, crimp the end with crucible tongs to seal it off?allow it to cool som ewhat before opening the tongs. Re-heat the tubing at the loosed end by passing it in and out of the flame until 1-2 inches of the tubing is in the molten state (looks transparent instead of opaque).Place the molten tubing into a small wide-mouth Jar or vial. Blow air into the open end of the tube so that it expands within the Jar. Let the expanded tubing cool in the Jar for about 2 minutes. Because the polymer is flexible, you should now be able to easily remove it from the â€Å"mold†. Show your polyethylene bottle to your TA. Polymer properties Supersaturates Weigh 1 Goff sodium polymerase and place it in a 150 ml beaker. Add 50 ml of water to the beaker from a graduated cylinder. Record your observations.
